안녕하세요 선생님, 강의 너무 재밌게 듣고 있습니다. 

강의에서 나온 두번째 실험에 관해 질문이 있습니다. Chloroplast in acidic environment --> alkaline solution was added --> ATP concentration was measured 이 실험입니다. 


You explained that because the proton concentration in the thylakoid lumen is similar to that in the stroma, no more ATP can be created because of the absence of a concentration gradient. I understand that explaination, but I just had one skepticism regarding the continuous production of protons though photolysis. If protons can be produced with the presence of H2O, wouldn't the concentration gradient still be present? Or, is the proton production from photolysis so insignificant compared to the proton yield from the proton pumps?

Excellent question! I love that you are thinking critically about the concepts while listening!

 

Short answer, yes.

 

The amount of protons produced from photolysis will be relatively insignificant in the overall picture.

We are talking about acidic (pH 4) and alkaline (pH 8) solution differences here.

The overall proton concentration difference is 10,000x higher in acidic solutions.

The concentration gradient formed by photolysis will not be a significant factor in this short snippet of time used for the experiment.

 

More importantly though,

read the question again carefully!

the first statement specifies that the chloroplasts are placed in a beaker "inside of a dark room"

 

what is the most important thing you need for "photo"lysis to occur?

That statement was designed specifically to answer questions from smart cookies like you!
